Specimen: | Bone Marrow, blood, lymph node, spleen, bone core, or effusion.
It is best if bone marrow specimens are obtained Monday through Sunday from 7:00 am to 3:00 pm. After 3:00 pm, bone marrows must be scheduled in order to provide coverage. Patient should not be on chemotherapy. |
Volume: | 2 mL bone marrow, 3 mL peripheral blood, or 0.5-1 mL tissue biopsy. |

Collection: | Bone marrow or peripheral blood in pediatric Green (heparin) tube; lymph node, spleen, bone core, CSF, or effusion in lymph node transport bottle.
Specimens should arrive in the testing laboratory within 48 hours of collection.
Maintain specimen at room temperature.

Rejection Criteria: | Hemolysis; gross contamination; no immature cells; clotted specimen; frozen specimen; fixed specimen; use of incorrect or too much anticoagulant; improper labeling. |

Methodology: | Short-term cell culture; synchronization; chromosome harvest; G-banding; analysis (20 metaphases) and karyotyping; long-term β-mitogen cultures in some B-cell leukemias. |
Test Use: | Identify specific chromosome abnormalities associated with chronic and acute leukemia and lymphoma, as well as myelodysplasia. |
Additional Information: | It is best if bone marrow specimens are obtained Monday through Sunday from 7:00 am to 2:30 pm. After 5:00 pm, bone marrows must be scheduled in order to provide coverage. Patient should not be on chemotherapy. |
